抗体薬物複合体の開発はMilsteinとKohlerのハイブリドーマ法の開発に端を発し、1970年代から行われており、当初はミサイル治療と呼ばれ、大変期待されたが、マウス抗体であったこと、抗腫瘍剤と抗体を結合するリンカーテクノロジーが未熟であったことなどから、臨床応用されることはなかった。その後、抗体工学の進歩により、キメラ抗体、ヒト化抗体、完全ヒト抗体などの作製技術が確立したことに加え、リンカーテクノロジーの進歩もあり、ADCは血液系腫瘍を対象として臨床開発が進んでいたが、2013年にTDM-1が転移性乳がんにFDAにより承認され、ADCの一般固形がんに対するはじめての本格的参入ということで、大きなインパクトを与えた。実際、2015年のアメリカ腫瘍学会(ASCO)におけるFDAの発表によると、現在のADCの治験は優に50を越える数にのぼっている。最も、そのほとんどが、アメリカ発のADCとなっており、ADCにおいても、日本はアメリカにほぼ完全に負けている。このままでは、さらに医薬品の輸入超過に拍車を掛けることになるであろう。

このような状況下で、色んなパーツの複合体といえるADCの本邦における成功をめざし、アカデミアと産業界から、それぞれの分野の第一線の研究者の方々に執筆していただいた。本書が、ADCに興味をもっていただくための参考書となることを祈念している。

<p>
2013年に「ブロック共重合体の自己組織化技術の基礎と応用」(以下「ブロック」)というタイトルでブロック共重合体の自己組織化によって形成されるモルフォロジーとその応用に関して、基礎的な部分から最新情報も含めた知見をこの分野の第一人者の諸先生方に執筆をお願いして、体系的にまとめたものを出版した。この当時、ブロック共重合体の自己組織化によって形成されるボトムアップ型の長距離秩序を持ったナノ構造は、ナノテクノロジーへの応用の期待されていたため、2012年におけるブロック共重合体関連の論文数は3700報にも上っており、その関心の高さもあって、「ブロック」も好評であった。<br />
「ブロック」を出版して以来5年がたつが、昨年1年間でのブロック共重合体関連の論文数は2012年からさらに増えて4500報になっており、ブロック共重合体に対する注目度はいまだに高い。<br />
その様な状況を鑑みて、「ブロック」に続くものとしてこの本の企画がなされた。この本においては、ブロック共重合体の形成する準結晶や、反射率測定など新しい物理・解析法、様々なブロック共重合体の新規合成法、新しいブロック共重合体の応用など、「ブロック」以降の新しい展開を網羅している。この本が「ブロック」同様、大学、研究機関、企業でのブロック共重合体の研究・開発に従事されていらっしゃる方のお役に立てれば幸いである。<br />
